Inhibitory effect of TRF2 on damaged telomere C-NHEJ. (A) The damaged shortened telomere ends bind to the Ku70/80 protein, and via mutual affinity of the Ku70 5α domains, a Ku70/80 heterotetramer is formed, leading to chromosome end-to-end fusion. (B) TRF2 directly binds to the 5α domain of Ku70 to prevent the formation of the Ku70/80 heterotetramers and inhibits the recruitment to the telomere ends of DNA-PKcs, Lig4, XRCC4, XLF, making the ends unable to be effectively connected.
