PfAP2-O-GFP-HA-DD24 is expressed in schizont stage parasites. (A) Fluorescence microscopy of late schizont parasites shows that GFP-tagged PfAP2-O colocalizes with the DAPI (nuclear stain) signal. WGA marks the surface of red blood cells. (B) Western blot of cytoplasmic and nuclear fractions obtained from PfAP2-O-GFP-HA-DD24 expressing parasites shows that AP2-O-GFP-HA-DD24 is found exclusively in the nucleus of schizonts. The membrane containing the transferred proteins was cut in three parts and was probed with monoclonal anti-HA (upper part), polyclonal anti-TPK (middle part), and monoclonal anti-Histone 3 antibodies (lower part).
