Silencing of SIRT3 prevents ANXA1sp-mediated protection from kidney cell death. (A) HK-2 cells were exposed to normoxic conditions or oxygen-glucose deprivation (Hypoxia) for 12 h and mRNA levels of SIRT3 were determined by RT-PCR. Graph displays mean +/- SEM (n = 3/group) with statistical significance determined by unpaired T test (*p < 0.05). (B) HK-2 cells were treated with control (-) or SIRT3 (+) siRNA. Levels of SIRT3 mRNA were determined by RT-PCR. Graph displays mean +/- SEM (n = 3/group) with statistical significance determined by unpaired T-test (***p < 0.001). (C) HK-2 were treated with control or SIRT3 siRNA. Cells were then pre-treated with Vehicle or ANXA1sp peptide for 1 h prior to oxygen-glucose deprivation for 16 h. Shown is fold change in cell death over Vehicle-treated, control siRNA cells. The protective effects of ANXA1sp were abrogated by SIRT3 siRNA (n = 6/condition).
