(a–d) MSCs characterization after in vitro expansion. (a) Cells, isolated from the bone marrow of a healthy donor, display the typical spindle-shaped morphology of MSCs. (b) Immunophenotyping by flow cytometry reveals expression of MSCs markers (CD73, CD90, and CD105) and lack of hematopoietic markers (CD34, CD45, CD80, CD86, and HLA-DR). (c) After incubation with the appropriate medium, the isolated cells were able to differentiate into osteoblasts as revealed by Alizarin Red staining (Sigma-Aldrich), which highlights calcium depositions. (d) At the same time, the isolated cells were also able to differentiate into adipocytes, as detected by Oil Red O (Sigma-Aldrich), which makes fat droplets visible.
