A fluorescence polarization assay with the fluorescently labeled PMCA peptide as probe was performed. For comparison Kd(OphA) = 3.5 ± 0.2 μM. While some compounds showed faint autofluorescence under the polarization assay conditions, their emission was too weak as compared to that of fluorescein to interfere with the measurements (Supplementary Figures 2C,D).
