CS-semi5 inhibits activation of the NF‐κB pathway. (A) RAW264.7 cells were treated with CS-semi5 (0.01, 0.1 or 1 μmol/L) for 24 h. IKK, IκBα, and NF‐κB phosphorylation was examined by western blot. (B) LPS-induced nuclear translocation of NF‐κB p65 was assessed by confocal laser fluorescence microscopy. Cells were immuno-stained with an NF‐κB p65 antibody (FITC, green), and nuclei were stained with DAPI in blue. Bar indicates 10 μm. (C) 293T cells were transfected with NF‐κB luciferase plasmid, then treated with CS-semi5 for 24 h. Luciferase activity was assayed. (D,E) RAW264.7 cells were stimulated with LPS and treated with CS-semi5 (0.01, 0.1 or 1 μmol/L) for 24 h.
