Association of PF14-SCO nanoparticles carrying different protein corona with cells. Complexes of PF14 with Cy5-SCO prepared at molar ratio 5, were incubated with HeLa cells in culture medium containing 10% of foetal bovine serum (a), murine serum (b), or human serum (c), or lacking serum (d) for 1 h. Lysosomes and other acidic organelles were visualised with Lysosensor Green. Localisation of nanoparticles based on Cy5 label on SCO (red), and acidic organelles (green) was captured in living cells with confocal microscope FV1000 and cell-associated SCO-Cy5 was quantified using Autoquant software (e). Scale bar 30 µm. Representative images from three independent experiments are presented. In panel (e), bars represent average fluorescence intensity per cell ±SEM. Data was analysed using one-way ANOVA with post-hoc Tukey’s test. **** p < 0.0001.
