Oligomerisation of EspB is promoted by an acidic environment. (A) Size exclusion chromatography profiles of M. tuberculosis EspB2-358 at 210 ​μM in 20 ​mM acetate buffer (pH 5.5), 150 ​mM NaCl and 20 ​mM Tris (pH 8.0), 150 ​mM NaCl. Void volume corresponds to 0.8 ​mL elution volume. (B) Oligomer/monomer ratios at different protein concentrations in conditions from panel (A). The absorbance values of the oligomer were taken at 1.14 ​mL while the monomer values were at 1.48 ​mL. (C–D) Presence of the different oligomer species from M. tuberculosis EspB2-348 at pH 5.5 and pH 8.5 obtained by native mass spectrometry.
