A computer model was developed to simulate delivery kinetics of IV-DDS. The model was validated by quantitative intravital microscopy studies where fluorescent dye encapsulated in thermosensitive liposomes (TSL) served as example IV-DDS. The computer model was based on in vitro measured IV-DDS (i.e., TSL) release kinetics, and on in vivo measured parameters from the in vivo studies with free dye (parameters marked by asterisks (*), see Fig. 6). The computer model predicted intra- and interstitial/EES concentrations during IV-DDS delivery, which were validated against results from the intravital studies (right-side graphs).
