Dlg1 deficiency impairs the capacity of Ag uptake by DCs. (A) Ag uptake by WT or Dlg1-KO splenic cDCs. Purified splenic cDCs were incubated with AF647-OVA at 0 or 37°C for 30 min. After washing with cold PBS, cells were measured for median fluorescence intensity (MFI) of AF647. Representative data from three independent experiments were shown (one mouse per genotype assayed in at least triplicate wells). (B and C) Representative flow cytometric graphs (upper panels) and quantification (bottom panels) of phagocytosis of WT and Dlg1-KO BMDCs. BMDCs were incubated with IgG-coated latex beads in the absence (B) or presence of 10 μM cytochalasin D (C). Percentages were calculated by the phagocytosed CD11c+ BMDCs in CD11c+ BMDCs.
