Lactate reduces the expression of MLH1 via GPR81-cAMP-PKA-CREB axis. SW480 cells were transfected with GPR81 siRNA and were then treated with or without lactate. (A) Protein expression of MLH1 and MSH2 was detected by Western blot. (B) mRNA expression of MLH1 and MSH2 was detected by qPCR. (C) Intracellular cAMP concentration was determined by ELISA. (D) Immunofluorescence indicated the nucleus translocation of PKACα cells. (E-F) Protein expression of p-CREB in nuclear was detected by western blot and immunofluorescence. (G) SW480 cells were treated with lactate in the presence or absence of Forskolin. Protein expression of p-CREB in nuclear was detected by western blot.
