(A) Mdm2/X binding site analysis by HSQC NMR. Chemical shift changes were observed for Mdm2/X residues in the primary (orange) and secondary (green) binding pockets upon addition of 1 eq of 4HBS. (B) The difference in chemical shift perturbation caused by 4HBS and 3HBS reveals that 4HBS engages the secondary binding pocket. Full HSQC spectra are included in the Supporting Information.
