Cell viability assay was performed to assess the effect of PEITC on HaCaT and CaSki cell lines. (A) PEITC exerted insignificant cytotoxic effects on HaCaT cells at various doses. (B) Morphological analysis showed that phenethyl isothiocyanate at doses of 20, 25, and 30 μM did not significantly alter the morphology of HaCaT 24 h after treatment. (C) Represents the structure of phenethyl isothiocyanate. (D) Phenethyl isothiocyanate resulted in significant cytotoxic effects in CaSki cells after 24 h of treatment in a dose-dependent manner when compared to the untreated control. (E) Cytotoxic effect of phenethyl isothiocyanate as analyzed by phase contrast microscopy. Similar to the cell viability result, phenethyl isothiocyanate sufficiently induced morphological changes in CaSki cells at 20, 25, and 30 μM after 24 h of treatment. (F) Inhibition of cell survival after 48 h of treatment with phenethyl isothiocyanate further increased as compared to that after 24 h in CaSki cells.
