Osteoarthritis‐associated catabolic factors up‐regulate subsets of Toll‐like receptor (TLR) signaling components in chondrocytes. A, Chondrocytes were treated with interleukin‐1β (IL‐1β) (1 ng/ml) or infected with 800 multiplicities of infection of adenoviruses Ad‐HIF‐2α or Ad‐ZIP8. Vehicle and control adenovirus were used as controls. Heatmap shows expression of TLR signaling components in the chondrocytes after treatment. B and C, Primary‐culture chondrocytes were treated for 36 hours with IL‐1β (1 ng/ml), IL‐6 (100 ng/ml), lipopolysaccharide (LPS) (10 ng/ml), or 800 multiplicities of infection of control adenovirus, Ad‐HIF‐2α, or Ad‐ZIP8. Levels of mRNA for the TLR‐2 and TLR‐4 genes (B) as well as for other genes (C) were determined by quantitative reverse transcription–polymerase chain reaction analysis (n = 6 per group).
