Schematic representation of the dual luciferase assay used to quantify inhibition of interferon signaling by Zaire Ebolavirus viral protein 24 (VP24) and to screen for inhibitors of this interaction. Cells were seeded and cotransfected with VP24 and pISRE-luc. The cells were stimulated and then lysed to measure luciferase activity. The expression of VP24 led to an inhibition of ISRE and a weaker luciferase signal. Effective inhibitors of VP24 could be added at the stimulation step, and the cells will thus produce a stronger signal.
