(A,B) C2C12 myotubes or isolated mouse skeletal muscle (extensor digitorum longus) were pre-incubated for 30 min with vehicle (0.1% DMSO) or the indicated concentrations of SBI-0206965 followed by an additional 1-h incubation without (vehicle) or with AICAR (1 mM). Cell or tissue extracts were subjected to ZMP analysis by liquid chromatography-mass spectrometry. (C) C2C12 myotubes were incubated in the absence (vehicle) or presence of the indicated compounds for 30 min followed by an addition of [³H]-uridine for 1 min for determination of uridine uptake.
