Cyclic peptomers inhibit the Salmonella SPI-1 T3SS. Salmonella enterica Typhimurium was grown with increasing concentrations of cyclic peptomer isomers. Secretion of SPI-1 T3SS cargo into the culture supernatant was assessed by precipitating secreted proteins and visualizing them with Coomassie blue. SipA and SipC band intensities were quantified and normalized to that of the DMSO control. (A and B) The experiments were carried out without the detergent Tween 20 (A) or with Tween 20 (B). A ΔSPI-1 Salmonella mutant and INP0007, a known SPI-1 inhibitor (64), were used as controls. The results are from at least two independent experiments. Error bars are standard errors of the mean.
