Examples of CD44/CD62L flow cytometry plots after vehicle or UNC2025 treatment. Data quantified in Fig. 7 A. WT NOD female mice were treated with 30 mg/kg UNC2025 or vehicle twice by oral gavage. Islets and LNs were isolated 17h after treatment initiation, digested, stained and analyzed by flow cytometry. Gating of islet samples was done independently from gating of LN samples due to partial cleavage of CD44 by Collagenase P, which was used for the islet isolation. Previous tests treating split samples of splenocytes with Collagenase P under the conditions used for islet isolation compared with no Collagenase P showed diminished florescence intensity of CD44 but equal frequencies when gating was adjusted. CD44 gates were set for islet samples and LN samples using CD62L+ cells as a CD44 population.
