Constitutive β-cell–specific deletion of CIC does not alter β-cell response to diet-induced obesity. Male CICfl/fl; Ins1-Cre vs. CICfl/fl mice were maintained on a Western diet (40% fat/kcal, 43% carbohydrate) for up to 12 weeks. A and B: Following 8 weeks of diet, 4–6-h fasted mice were injected with glucose 1 mg/g body wt i.p. (n = 11–16). Blood glucose (A) and plasma insulin (B) were recorded at the indicated times. C: Following 8 weeks of diet, isolated islets were perifused for 32 min at basal (2.5 mmol/L) glucose and then stimulatory glucose (11.2 mmol/L) for the indicated times (n = 6).
