Two potential methods for using ECCITE-seq to determine eQTL function. Mixed neural cultures constitutively expressing CRISPR effectors are transduced with lentiviral guideRNA libraries targeting known eQTL genes. ECCITE-seq can be subsequently used to concatenate global gene expression data, cell identity data and guideRNA expression data in individual cells. Cultures infected with guideRNA libraries at a low MOI can be used to determine the impact of individual perturbations, while high MOI cultures can be used to investigate combinatorial perturbations and synergistic effects.
