ScHSF binds cooperatively to its target HSEs on naked hsp82 templates. Linear DNA templates (1 nM), amplified by PCR and bearing the HSP82⁺ or hsp82-P2 upstream sequence, were titrated with increasing amounts of GST-ScHSF (0, 0.4, 1.2, 3.6, 33, and 100 nM in lanes 4 to 9, respectively). Resultant protein-DNA complexes were digested with DNase I, and the digestion products were processed, electrophoretically separated on an 8% sequencing gel, and visualized with a PhosphorImager.
