GTPases are required for ROI production in response to IL-1β in monocytic cells. U937 cells were transfected with the CD20 expression vector and with plasmids coding for wild-type (Cdc42 and Rac) or dominant-negative (Cdc42^m and Rac^m) Rac1 and Cdc42. CD20-expressing transfected cells were positively selected with an anti-CD20 monoclonal antibody through MACS and flow cytometry cell sorting. Selected cells were either untreated (−) or stimulated with IL-1β (+) for 15 min, and formation of ROIs was measured by using a DFCH probe.
