Induction of COX-2 signaling by MAI is dependent on miR-155 in macrophages. BMDM were transfected with antagomir against miR-155 and control. Then, cells were infected with MAI at MOI of 25. (A) Expression of COX-2 mRNA was detected through RT-qPCR. (B) Cell lysates were immunoblotted with anti-COX-2 antibody, and Tubulin as the internal control. (C) Concentration of PGE2 in the supernatant of cells was evaluated through ELISA. Binding of Ago2 with miR-155 (D) and COX-2 mRNA (E) was detected by RNA immunoprecipitation (RIP) using anti-Ago2 and RT-qPCR.
