Ctk2p turnover in early exponential cells. Δctk2 cells transformed with the CTK2-HA construct were grown in SD medium to an OD₆₀₀ of 0.8. Aliquots were taken at different times (minutes) of incubation at 30°C after the addition of cycloheximide. Resulting cellular extracts were analyzed by Western immunoblotting. After the addition of cycloheximide, Ctk2p levels were monitored for 180 min (A) and 5 min (B) to analyze turnover of the nonphosphorylated and phosphorylated species, respectively.
