Human PBMCs express anti-inflammatory signatures in response to cGAMP MPs. (A) Monocytes isolated from PBMCs from RRMS patients were treated with 5 ug/ml cGAMP MPs for 8 h and evaluated for IFN-I gene expression by quantitative RT-PCR (qRT-PCR). (B) RRMS patient PBMCs were treated with cGAMP MPs for 24 h and stimulated with LPS prior to monocyte staining for flow cytometry. (C) Monocytes from RRMS patients were cultured with cGAMP MPs for 24 h, and supernatants were collected for ELISA. (D) RRMS patient monocytes were stimulated with LPS and pretreated with cGAMP MPs for 12 h. Washed monocytes were cocultured with autologous CD4+T cells for 12 h. Treatment groups were compared by a one-way ANOVA. Data are combined of at least two independent experiments (n = 4–7).
