Engineered synthases and their productivities in vivo. Ethyl acetate extracts of culture broths were analyzed by HPLC with a C18-column. Diamonds indicate triketide lactone peaks. a) The 1-polypeptide Pik127, 2-polypeptide Pik127, and Pik167 were designed with the updated and traditional module boundaries (left and right, respectively). Those designed with the updated boundaries were >10-, 3.3-, and 2-fold more active, with the updated Pik167 producing 0.39 g L-1. Cultivations were performed in triplicate (measurements and standard deviations in Source Data 2), and productivity was calculated from peak areas using a standard curve (Figure S3, Source Data 1).
