A) In wildtype epithelial cells, Scrib forms a complex with Lgl and Dlg that antagonizes the aPKC/Par6/Par3 complex to establish the basolateral domain and target MIIB to the junctions leading to the stabilization of cell junctions. B) Loss of Scrib frees Lgl allowing Lgl to bind with aPKC/Par6 and displace Par3 from the complex destabilizing junctions. Furthermore, ZO-1, DLC3, and MIIB are no longer recruited to the junctions by Scrib, instead MIIA is targeted to the junctions by Lgl.
