Upon binding of the Notch ligand to the receptor, the Notch intracellular domain (ICD) is cleaved from the extracellular domain (ECD) and translocates to the nucleus where it binds to the downstream transcription complex and activates target gene expression. Notch activation induces differentiation of limbal stem/progenitor cells (LSCs) and decreases proliferation and stratification. Notch signaling inhibition in LSCs by small molecule inhibitors maintains LSC phenotype and induces proliferation and stratification.
