Effect of AdoMet and/or 5-FU on the levels of P-gp, NF-κB, and its acetylated form in HCT 116p53+/+ and LoVo cells. Cells were treated with 5-FU 50 µM for 24 h, with and without pretreatment with 500 µM AdoMet for 48 h. Total RNA was extracted and cDNA was synthesized by qRT-PCR. The graphs show the mRNA levels of P-gp in HCT 116p53+/+ (A) and LoVo (B) cells normalized to GAPDH mRNA. Data represent the average of three independent experiments. Western blot assay was used for the expression of NF-κB and acetylated NF-κB in HCT 116p53+/+ (C) and LoVo (D) cells, respectively. β-actin was used as loading control. Graphs show the relative densitometric analyses, expressed as percentage of untreated control (100%). Error bars represent the standard deviation, * p < 0.05 versus untreated cells.
