The RXL motif in the COOH terminus of Myt1 is required for cyclin B1 binding. HeLa cells were transiently transfected with empty vector (control) (lane 1) or vectors encoding either Myc epitope-tagged Myt1 (mycMyt1) (lane 2) or Myc epitope-tagged Myt1 containing alanines in place of RNL at positions 486 to 488 [mycMyt1(3A)] (lane 3). Myt1 proteins were immunoprecipitated with antibody specific to the Myc epitope, and precipitates were analyzed for cyclin B1 (top panel) and Myt1 (bottom panel) by Western blotting.
