Neither Sir3p (A) nor Sir4p (B) is involved in the repression of RP gene transcription in a sec mutant or during heat shock. Strains of the indicated genotypes (Table 1) were grown to log phase in YPD at 23°C. An aliquot was harvested, the cultures were shifted to 37°C, and aliquots were harvested at the indicated times. RNA was prepared and analyzed as described previously, except that all RNAs were detected with ³²P-labelled oligonucleotide probes.
