TG causes mitochondrial depolarization that is inhibited by Bcl-2 or Bcl-XL but not by the caspase inhibitor z-DEVD-fmk. (A) JT/Neo, JT/Bcl-2, and JT/Bcl-XL cells were treated with TG (100 nM) for various times or left untreated. During the last 30 min of treatment, DiOC6(3) was added. An aliquot of the cells was used for the determination of cell-associated DiOC6(3) fluorescence. (B) Jurkat cells were pretreated for 30 min with z-DEVD-fmk before addition of 100 nM TG, and uptake of DiOC6(3) was determined at the times indicated.
