Pyrene-actin assembly assays to compare the nucleation activity of 5 nM WT or mutant FFCs with (A) 2 μM actin or (D) 2 μM actin and 4 μM S. pombe profilin. (B) Polymerization rate at 50 seconds and (C) time until half-maximal polymerization (thalf) for pyrene-actin assay with 2 μM actin alone at 0, 1, 2 and 5 nM concentrations of FFC. (E) Polymerization rate at thalf and (F) thalf for 2 μM actin and 4 μM S. pombe profilin at 0, 1, 2 and 5 nM concentrations of FFC. In all assays, 5% of the actin monomers were pyrene-labeled.
