Comparison of TG cells from 2DG treated and untreated HSV infected mice at day 9 pi for T cell activation and IFN-γ producing function. Infiltrating T cells were isolated from cultures after enzymatic dissociation of 8 individual TGs. T cells were stimulated with PMA/Ionomycin and surface stained with CD4, CD8 and CD69 fluorochrome-labeled antibodies. Cells were also reacted with anti- IFN-γ to show intracellular IFN-γ as a measure of function. Panel A and B shows CD69+CD8+ differences between HSV infected untreated and 2DG treated groups. CD69 expressing total CD4 T cells were also measured (C) and the FACS plots present the percent changes within CD4 T cells (D). IFN-γ producers measured (E) and gated from activated CD8 T cells (F). IFN-γ expressing CD4 T cells were measured (G) after gating CD69+CD4+ T cells and presented with FACS plots (H). A Student t-test was used to measure statistical significant differences between treated and untreated groups (**p<0.01; ***p<0.001; ****p<0.0001).
