ASCs in transwell culture downregulate profibrotic gene expression in irradiated fibroblasts. (A): Schematic of transwell coculture. (B) Human ASCs were cocultured with nonirradiated or 10 Gy irHFF and tested at 48 hours post coculture. Fibrosis-related gene transcripts for TNF, NF-κB, IL-1, CTGF, TGF-β, Col1, Col4, and Col5 were measured by real-time polymerase chain reaction (RT-PCR) in the HFF cells. Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) was used as a housekeeping gene (n = 3). (C): Mouse bone marrow stromal cells were irradiated to 10 Gy and cocultured in transwells below mouse ASCs for 48 hours. mRNA expression of profibrosis genes TNF, NF-κB, IL-1, CTGF, TGF-β, Col1, Col4, and Col5 was measured by RT-PCR (n = 3).
