(A) In health, pericyte insulin signaling regulates Ang1 secretion and hence pericyte-endothelial crosstalk via Ang1-Tie2, promoting endothelial FoxO1 nuclear exclusion and preventing Ang2 transcription. (B) In PIRKO, insulin receptor signaling is disrupted, which reduces Ang1 secretion and downstream activation of endothelial Tie2. The transcription factor FoxO1 remains nuclear and allows transcription of Ang2.
