Identification of the specific cleavage site of mRNA delivered by RNA nanoparticles into KB cells

(A) Site-specific cleavage of Lamin A/C mRNA induced by pRNA-lamin demonstrated by 5′ RACE. pRNA-lamin25 was constructed by linking a siRNA sequence (underlined) with a pRNA vector via UU linkers at the sense (upper) and antisense (lower) strand. Sequences of Lamin A/C mRNA that are identical with the siRNA are underlined, and the cleavage site is marked with a star. Sequencing results show that the PCR product contains a RNA ligator sequence (red) and part of the Lamin A/C mRNA sequence. (B) PCR products resulting from outer primer pairs were separated in 1.2% agarose gel.
