(a) Puf3p binds to the long FAD1 mRNA and drives the nascent polypeptide in the mitochondrial matrix (MMX) via TOM–TIM complexes before the fully translated protein starts to fold. (b) Puf3p binds to the short FAD1 mRNA. As translation is presumably not sufficiently slowed down, premature and rapid folding of the nascent polypeptide in the cytosol (CYT) prevents Fad1p import across mitochondrial membranes (OMM—outer mitochondrial membrane; IMM—inner mitochondrial membrane).
