IL-1 treatment does not reduce NSC self-renewal or deplete NSCs in neonatal mouse hippocampal NP cultures. NPs derived from the hippocampi of neonatal C57BL/6 or Swiss Webster mice were grown in the presence of IL-1 for 7 days and either passaged repeatedly from three weeks or seeded into plates for limiting dilution analysis (LDA). (A) Quantification of cell yield after each passage of NPs that were either maintained in standard growth medium or treated with 500 pg/mL IL-1β. Results are expressed as mean ± SEM; ∗p < 0.05 by unpaired t-test, n = 3 independent experiments. (B) Representative graph depicting sphere-forming frequency of control versus 500 pg/mL IL-1β-treated NPs in LDA. (C) Timeline of treatment and protocol for repeated passaging and LDA.
