IDH activity in hNs undergoing nitrosative stress hNs manifest inhibition of isocitrate dehydrogenase (IDH) enzyme activity in their mitochondria due to S-nitrosylation. Wild-type (WT) hiPSC-derived neurons (hNs) were exposed to 100 μM of the physiological NO donor S-nitrosocysteine (SNOC), and the lysates analyzed for IDH activity with isocitrate as substrate in the presence of NADPH with NADP+ being the assayed species reflective of IDH activity. A SpectraMax microplate instrument was used for the measurements. Values are mean + SEM. p < 0.05 by Student’s t test; n = 3.
