Levels of cellular ceramide were quantified from primary myoblasts in culture. Cells from the WT (at passage 8 in vitro) and VCP^R155H/R155H (HZ, at passage 7) mice were cultured and scraped into methanol. The lipids were extracted with chloroform and the levels of various ceramide species were determined by using LC–MS/MS (n = 5–6).
