Unmodified vesicles can be profiled with RDIMIS using cholesterol as a readout. (A) PD-L1 gD-GPI or PVR gD-GPI rEVs containing gag-RLuc were diluted across a range of concentrations and then read out either by luciferase signal or using the AmplexRed Cholesterol Assay to measure cholesterol content of the solutions. (B) Small-batch RDIMIS with PD-L1 gD-GPI rEVs was performed and read out using either cholesterol or luciferase signal to compare the signal in a screening context.
