Gpc4 is required for the formation of Wnt5b-labeled protrusions in endodermal cells. Confocal time-lapse experiments were performed on gpc4−/−/Tg(sox17:memGFP)/ Tg(sox17:H2AmCherry) embryos injected with Wnt5b-mCherry using a Zeiss LSM880 confocal microscope with a LD C-Apo 40×/NA 1.1 water objective (shown in Fig. 7, C and D). Z-stacks of 13.5 µm were acquired at 1.5-µm intervals every 30 s using the following settings: zoom 1.0, 1,024 × 1,024 pixels, 9 speed, 4 averaging.
