Primer extension occurs via two sequential reactions. (A) Formation of the imidazolium-bridged dinucleotide intermediate, the reactive species in nonenzymatic RNA primer extension. (B) The imidazolium-bridged intermediate anneals to the primer-template duplex and reacts with the 3′-end of the primer.
