For RT-PCR assays, cDNA reactions were prepared by using total RNAs isolated from unpurified thymocytes of wild-type and transgenic littermates at the indicated ages. PCR products were analyzed by Southern blotting with radiolabeled probes and quantified with a PhosphorImager.

The amount of each listed PCR product was normalized to that of GAPDH. The value for the transgenic mice was then divided by that for their wild-type littermates and multiplied by 100. The Vβ transcript was not detectable in 1-week-old transgenic mice; it is therefore listed as 0.
