a Analysis of ATPase activity in sorted T cell subsets of two donors by HPLC. Activated cells (αCD3/αCD28, 3 days) were incubated with 1,N6-etheno-ATP (eATP) and degradation of etheno-nucleotides was determined by HPLC. Histograms show CD39 expression on the cells at the time of eATP incubation. b CD73 CD4con T cells were stimulated with αCD3/αCD28 in the presence of the ADA inhibitor EHNA (10 µM) and incubated with Tregs, ATP (50 µM), and EVs from cell culture supernatants of sorted and activated CD8 T cells as indicated. CD25 expression and proliferation were measured after 4 days by flow cytometry. IFNγ production was determined by ELISA in the cell culture supernatant harvested on day 4. Data were shown for one representative donor out of four analyzed (mean of technical replicates).
