Cytotoxic and hemolytic activity of peptides. (A) Cytotoxicity of DIII blocking peptides assessed by XTT test after 24 h of incubation at different concentrations. A statistically significant difference (P < 0.01, two-tailed P-value) was calculated by unpaired t-test with Welch´s correction compared to the negative control (untreated cells). None of the peptides showed significant cytotoxicity. Statistics was performed with GraphPad Prism v8.4.3. “-” – negative control (untreated hBMECs); “+” – positive control (cells treated with 0.1% Triton X-100); CP – 7-mer cyclic peptide. (B) Assessment of hemolytic activity of peptides. Suspension of sheep erythrocytes was incubated with peptides at two different concentrations.
