Real-time quaking-induced conversion of α-synuclein. (A) Outline of RT-QuIC. Tissues or body fluids of patients are added to a reaction buffer that contains recombinant α-synuclein (rec α-syn). Aggregated α-syn converts rec α-syn to the abnormal structure, which forms amyloid by repeated shaking and incubation. Thioflavin T (ThT) binds to the β-sheet structure of amyloid and its fluorescence is detected. (B) Results of α-syn RT-QuIC.
