Application example: detection of cell killing by bortezomib. A FUCCI-C8161 melanoma spheroid was incubated with 50 nM bortezomib for 48 h and a central confocal fluorescence image was taken at 0 h (A), 24 h (B), and 48 h (C). DRAQ7 was added to the culture medium 24 h prior to bortezomib addition to a final concentration of 3 μM. Percentages of mAG⁺ (D–F), mKO2⁺ (G–I) and DRAQ7⁺ (J–L) cells were calculated using an image analysis similar to that applied for the example depicted in Figure 11 and plotted as a function of the distance from the spheroid surface.
