PLK1 is associated with gefitinib-induced liver damage. C57BL/6 mice were randomly divided into 4 groups (n = 6 per group). After injection of AAV8-shPlk1 adeno virus by tail vein for 3 weeks, mice were treated with CMC-Na or gefitinib (200 mg/kg/day) by gavage for another 4 weeks. (A) The levels of GPT/ALT and GOT1/AST were determined (n = 6, one-way ANOVA, Dunnett T3 test). (B) H&E staining, IHC for PLK1 and TUNEL staining of liver tissues. Scale bar: 100 μm. H&E staining distinguish normal or damaged sinusoidal structure. Red arrowheads indicated positive regions. (C) Liver tissues were extracted and lysed, and the expression levels of PLK1, COX6A1, LC3 and GAPDH were analyzed by western blot. Western blot was repeated at least three times and densitometric analysis was carried out. The data are expressed as the mean ± SD; *p < 0.05; **p < 0.01; ***p < 0.001.
